For fractioning of aminopeptidase activities, dialysed cheese or micro-organism extracts were submitted to batch chromatography in Q-Sepharose (Amersham), equilibrated in 20 mM sodium phosphate, pH 7.5, in a proportion of one A280 unit of protein per mL packed resin. Samples and resin were mixed in Becker and kept under stirring in an ice bath for 2 h. The mixture was poured into a glass-sintered funnel and washed with the equilibrating buffer to recover nonretained fractions. Three elution steps were carried out, adding NaCl to the equilibrating buffer: fraction I: 150 mM NaCl; fraction II: 300 mM NaCl; and fraction III 500 mM NaCl. The protein content and aminopeptidase activity of fractions were analysed as described.
