Another example of a dynamic culture system is the perfusion bioreactor. Such systems utilize a pump that allows for the medium to flow over culture surfaces. Incorporating flow into the culture conditions is used in both two-dimensional and three-dimensional cultures. For two-dimensional cultures, cells grow on an interior surface of a chamber, which has an inlet and outlet for medium to pass over the cells (Figure 17.11). The three-dimensional system is very similar, except that the medium either is pumped through the cell-containing scaffolds or is dripped on top of the scaffolds and allowed to flow through via gravity These bioreactors are usually closed systems, allowing for the medium to be passed over the culture surfaces several times before it’s replaced with fresh medium. Advantages to having the medium flow over the cells are realized through the effects that are brought about by having the cells exposed to controlled and directional shear, which aid in the differentiation of pluripotent and multipotent cells http://www.sciencedirect.com/topics/page/Bioreactors Once seeded, spheroids rest on a polycarbonate membrane with 12 µm pore size, allowing the medium to flow-through, while flow-over is maintained to avoid an excess pressure on the cells. https://biblio.ugent.be/publication/7003105 |
