Leggendo la frase precedente, ho il forte sospetto che il testo si riferisca sia alle estremità degli oligonucleotidi (3' e 5') sia alla posizione in cui i nucleotidi sono legati tra loro (posizioni 1, 2, 3, 4, come qui: http://www.chemgapedia.de/vsengine/vlu/vsc/en/ch/12/oc/vlu_o... ) Cercando, ho trovato questo brevetto, che tratta di oligonucleotidi modificati: https://patents.google.com/patent/WO2005115481A2/en Alla fine di pagina 22, dice: "Cleavage of RNA or dsRNA by nucleolytic enzymes requires the formation of an enzyme-substrate complex, i.e. a particular nuclease-oligonucleotide complex. The nucleases generally require specific binding sites on the oligonucleotide for appropriate attachment. If the binding sites are removed or blocked, such that the nucleases are unable to attach to the oligonucleotide, the oligonucleotide will become nuclease resistant. This concept is well established particularly for the protection of oligonucleotides from degradation of exonucleases, enzymes that degrade oligonucleotides exclusively from their ends. Per caso, l'uso di backbone modificati per questi primer ha lo scopo di aumentarne la stabilità? Penso che, quando il testo parla di "3' to 3', 5' to 5' or 2' to 2' linkages", si riferisca alla posizione dei legame tra nucleotidi (come qui https://www.biosyn.com/inverted-base-oligonucleotide-synthes... ), mentre quanto parla di "3'-most" si riferisca alla posizione del legame nella sequenza di nucleotidi (da 5' a 3'). -------------------------------------------------- Note added at 14 hrs (2018-10-15 09:46:12 GMT) -------------------------------------------------- Se è scritto "3'-most" (con l'apostrofo dopo il 3), secondo me sì. Nel contesto, l'alternativa sarebbe che l'espressione si riferisca ad un legame in posizione 3. Però questa ipotesi non ha molto senso. Se lo scopo è aumentare la specificità di un test (che immagino sia una PCR), l'uso di nucleotidi modificati nelle posizioni terminali dei primer usati per il protocollo B potrebbe diminuire la degradazione dei primer e quindi aiutare a confermare che i prodotti già ottenuti nel protocollo A riflettono effettivamente la presenza delle sequenze che si stavano cercando. Potrebbe avere senso. |
