the term "macrobead" refers to a structure that is essentially spherical, with a diameter of from about 4 to about 10-12 mm in diameter, most preferably from about 6 to about 8 mm in diameter. The second agarose layer is preferably from about 0.05 to about 5 mm in thickness, most preferably from about 0.5 mm to about 5 mm in thickness, even more preferably, from about 1.0 mm to about 3 mm, and most preferably, from about 1.0 mm to about 2.0 mm in thickness. The second agarose layer may, but need not be, the same agarose used to make the bead.
