The fasting-mimicking effects of metformin, a drug widely prescribed for type 2 diabetes, are mediated via modulation of the AMPK pathway, a central nutrient signaling pathway. We found that metformin-treated cells exhibited increased expression of Pdgfra and Ascl1 (Figures S4G and S4H), expressed significantly less Cdkn2a (Figure S4I), and had less DNA damage, as indicated by comet assays (Figure S4J), suggesting that metformin is sufficient to ameliorate some of the hallmarks of aging and phenocopy at least some of the effects of ADF.
