This assay is based in the biology of the horseshoe crab (Limulous). These animals produce LAL enzymes in blood cells (amoebocytes) to bind and inactivate endotoxin from invading bacteria. Проведение реакции не требует специального оборудования. Методика заключается в добавлении к предварительно растворённому LAL-реактиву исследуемого образца (напр., плазма крови, моча). Далее пробирку помещают на водяную баню и инкубируют 60 мин. при 37°С. Плотный гель на дне пробирки, не разрушающийся при ее переворачивании, называется положительной реакцией и означает, что концентрация эндотоксина в пробе больше или равна чувствительности LAL-реактива. Отсутствие плотного геля или его разрушение при переворачивании пробирки называется отрицательной реакцией и означает, что концентрация эндотоксина в данной пробе слегка или значительно ниже чувствительности LAL-реактива. |
