I can't find the referenced table so I don't know what's going on here. Is this double-character iroha or is the 二 a 2? If it's iroha would you use DE (or de)?

I think “二” is (No.) 2, and “ホ” is e (E as in Edward) in the Iroha (traditional ordering of the Japanese syllabary) order. For your information, ニ is Katakana, and 二 is the numeric character (2) in Kanji. And, the letter you have specified there is 二, which is obviously 2.

I searched 昭和54告示547別表二ホ, and now believe that ニ is 漢数字 of 2.
漢数字の二とカタカナのニは、良く似ています。
