The Seeplex® RV15 OneStep ACE Detection is a qualitative in vitrotest for the detection
of 15 respiratory pathogensin nasopharyngeal aspirate, nasopharyngeal swab, or
bronchoalveolar lavage specimens from symptomatic patients. Fifteen pathogens include
influenza A virus (Flu A), influenza B virus (Flu B), human respiratory syncytial virus A (RSV A),
human respiratory syncytial virus B (RSV B), human adenovirus (AdV), human
metapneumovirus (MPV), human coronavirus 229E/NL63 (229E/NL63), human coronavirus
OC43 (OC43), human parainfluenza virus 1 (PIV1), human parainfluenza virus 2 (PIV2), human parainfluenza virus 3 (PIV3), human parainfluenza virus 4 (PIV4), human rhinovirus A/B/C (HRV), human enterovirus (HEV), and human bocavirus 1/2/3/4 (HBoV).
Flu A: The primers detect all subtypes of influenza A virus including the H5N1 subtype (a highly pathogenic avian influenza) and the A(H1N1)pdm09 subtype (an influenza of swine origin which was first detected in April 2009).
AdV: The primers are designed to detect most of B, C, and E types and some of A and D types
of human adenovirus.
PRINCIPLES AND PROCEDURE OVERVIEW
1. Principles
The polymerase chain reaction (PCR) DNA amplification technique displays high sensitivity and specificity. Seegene developed a new concept of oligo technology called DPOTM (Dual Priming Oligonucleotide) which provides flexibility in primer design and PCR optimization and maximizes PCR specificity and sensitivity through the inhibition of non-specific priming.
The Seeplex® RV15 OneStep ACE Detection is a DPO TM -based multiplex assay that
simultaneously amplifies target sequences of influenza A virus (Flu A), influenza B virus (Flu B), human respiratory syncytial virus A (RSV A), human respiratory syncytial virus B (RSV B),
human adenovirus (AdV), human metapneumovirus (MPV), human coronavirus 229E/NL63 (229E/NL63), human coronavirus OC43 (OC43), human parainfluenza virus 1 (PIV1), human parainfluenza virus 2 (PIV2), human parainfluenza virus 3 (PIV3), human parainfluenza virus 4 (PIV4), human rhinovirus A/B/C (HRV), human enterovirus (HEV), human bocavirus 1/2/3/4 (HBoV), PCR control (PCRC), and whole process control (WPC).
Reverse transcription (RT) and DNA amplification (PCR) are set up together in a single tube in the Seeplex® RV15 OneStep ACE Detection.This simplification of the two-step
method results in high assay reproducibility and efficiency. The Seeplex® RV15 OneStep ACE
Detection is based on three major processes: nucleic acid isolation, target amplification by one-step RT-PCR using DPOTM primers, and detection on ScreenTape® System, MultiNA System, or an agarose gel electrophoresis.
The Seeplex® RV15 OneStep ACE Detection includes two internal controls: PCR control
and whole process control. The PCR control is added to both set A and B in order to check
for the presence of substances in the processed specimens that may interfere with PCR
amplification. Set C includes human RNase P target as an internal control to check the entire
experimental process from nucleic acid extraction to RT-PCR.
In addition, 8-methoxypsoralen (8-MOP) is usedto terminate the template activities of
contaminated DNAs. 8-MOP insets itself intodouble-stranded nucleic acids and forms a
covalent interstrand crosslink after photoactivation with 320-400 nm incidence light. Please discard used tubes after UV irradiation (365 nm) for 20 minutes to prevent carry-over
contaminations.

BASIC PRODUCT INFORMATION
Based on our vertical laminar flow cabinet product line, our PCR cabinet has been designed specifically for polymerase chain reaction (PCR) applications. The cabinet isolates critical steps in the PCR process by providing a particulate-free work environment and can be operated effectively as “mini-environments” in the laboratory. Vertical laminar flow cabinet offers proven protection for your sample and processes where operator protection is not required. They are suitable for application which requires ISO Class 3 air cleanliness within work zone as per ISO 14644-1 (equivalent to Class 1 per US Federal Standard 209E)
AIRFLOW PATTERN INSIDE THE CABINET
Room air is taken in from the top of the cabinet through a disposable pre-filter with 85% arrestance; this serves to trap larger particles and increase the life of the main filter. Air is forced evenly across an HEPA/ULPA-filtered vertical laminar flow air stream within the cabinet; the result is a stream of clean laminar air within the work zone of the cabinet; this dilutes and flushes all airborne contaminants from the interior. A nominal filter face velocity of 0.45 m/s or 90fpm ensures that there is sufficient number of air
changes within the enclosed are of the cabinet in order to maintain cleanliness. The purified air travels across the internal work zone of the cabinet in a vertical, unidirectional stream and leaves the main work chamber across the entire open front of the cabinet.
