Of note is AG10’s binding selectivity for TTR, which surpasses that of the natural ligand T4, and the completeness with which it excludes the fluorescent probe.
Another key consideration pertains to a TTR stabilizer’s ability to specifically interact with both wild-type and *variant TTRs*.
Variant TTR is less stable than wild-type and makes a more than proportional contribution to pathological amyloid deposition: when the composition of soluble plasma TTR is compared to TTR amyloid deposits in tissues of heterozygous ATTRm patients, variant TTR accounts for a third of the total TTR in plasma but within amyloid fibrils the proportion is reversed with two thirds being variant TTR (Dwulet 1984).
