Source text - English A similar procedure is described by Schmidt and
Poise1 (6) for the preparation of racemic proline.
The pyrrolidine 2a was obtained in five steps from
4-hydroxy-L-proline 3a.2 In order to obtain a
1-methylene-1-carbapenam with the same configuration
at C2 as in penicillins, 4-hydroxy-Dproline
should be used as a starting material.
However, in these experiments the more readily
available L-enantiomer was used. Oxidation of
N-tert-Boc-4-hydroxy-L-proline 3b with Cr0,lpyridine
in CH,Cl, gave the ketone 3c (60%), which
was converted into the N-protected 4-methylenepyrrolidine
3d by condensation with methylenetriphenylphosphorane
(4 equiv.) in THF (80% yield).

La pirrolidona 2a se obtuvo en 5 pasos a partir de la 4-hidroxi-L-prolina 3a. Para obtener 1-metilen-1-carbapenam con la misma configuracion en C2 como en las penicilinas, se debe emplear 4-hidroxi-D-prolina como sustancia de partida. Sin embargo, en estos experimentos se uso el enantiómero L por su fácil disponibilidad. La oxidación de N-tert-Boc-4-hidroxi-L-proline 3b con CrO3/piridina en CH2Cl, dio como resultado la cetona 3c (60%), la cual fue convertida en la 4-metilenpirrolidine protegida en el N 3d mediante condensación con metilentrifenilfosforane (4 equiv.) en THF (80% de rend.)

Source text - Spanish R [4-2H]-NADPH se preparó mediante una reducción estereoespecífica de NADP con 2-propanol-d8 (más del 99.7% D en 2C de acuerdo a la determinación por 1H-RMN) usando una alcohol-deshidrogenasa de Thermoanaerobium brockii (tbADH). Se sintetizó R [4-3H]-NADPH (680 mCi/mmol) mediante la reducción de NADP con glucosa-[1-3H] usando una glucosa deshidrogenasa (GluDH) de Cryptococcus uniguttulatus, seguida de una oxidación del NADPH resultante con acetona usando tbADH y posteriormente una segunda reducción con glucosa no marcada usando GluDH como se describe en mayor detalle en otras publicaciones. Se preparó [Ad-14C]-NADPH (50 mCi/mmol) mediante la fosrolilación en 2´ de [Ad-14C]-nicotinamida adenina dinucleótido (NAD ) usando una NAD kinasa de hígado de pollo para producir [Ad-14C]-NADP , seguida de una reducción con glucosa usando GluDH tal como se describe en otras publicaciones.
Translation - English R [4-2H]-NADPH was prepared through stereospecific reduction of NADP with 2-propanol-d8 (greater than 99.7% D at 2C as determined by 1H-NMR) using alcohol dehydrogenase from Thermoanaerobium brockii (TbADH). R [4-3H]-NADPH (680 mCi/mmol) was synthesized by reduction of NADP with [1-3H]-glucose using glucose dehydrogenase (GluDH) from Cryptococcus uniguttulatus, followed by oxidation of the resulting NADPH with acetone using tbADH and then a second reduction with unlabelled glucose using GluDH as described in more detail elsewhere.
[Ad-14C]-NADPH (50 mCi mmolK1) was prepared by 2´-phosphorylation of [Ad-14C]-nicotinamide adenine dinucleotide (NADC) using an NADC kinase from chicken liver to produce [Ad-14C]-NADP , followed by reduction with
glucose using GluDH as described elsewhere.
